چکیده

objective: hexavalent chromium [cr (vi)] compounds are well-known environmental contaminants  generated from industrial processes. several studies have reported the harmful effects of cr (vi) on different organs, however, little is known about neurotoxic effects of cr (vi). the aim of this study is to investigate the toxic effects of cr (vi) on pc12 cells. methods: pc12 cells were cultured following standard protocol and exposed to various concentrations (1-100 μm) of potassium dichromate (k2cr2o7) for 24, 48 and 72 h. after exposure, cell viability was measured by the mtt assy. after exposure, production of reactive oxygen species (ros) and lipid peroxidation were measured. results: potassium dichromate induced significant cell death in pc12 cells. the ic50 values for cytotoxicity were 22.02 for 24 h, 1.88 for 48 h, and 1.85 for 72 h of exposure. significant differences between ic50 for 24 h of exposure compared to 8 and 72 h of exposure were observed (p

Evaluation of Neuroprotective Effect of Althaea Officinalis Flower Aqueous and Methanolic Extracts against H2O2-Induced Oxidative Stress in PC12 Cells

Objective: This study was conducted to evaluate the possible antioxidant activity and neuroprotective effects of aqueous and methanolic extracts of Althaea officinalis flowers against H2O2-induced oxidative stress in PC12 cells. Material and Methods: The antioxidant potential of extracts was evaluated by radical scavenging activity using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. For c...

Berberine protects against glutamate-induced oxidative stress and apoptosis in PC12 and N2a cells

Objective(s): Neurodegenerative diseases have been associated with glutamatergic dysfunction. Berberine, an isoquinoline alkaloid broadly present in different medicinal herbs, has been reported to have neuroprotective effect. In the present study, the effects of berberine against glutamate-induced oxidative damage and apoptosis were investigated. Materials and Methods: The cultured PC12 and N2a...

Phlorofucofuroeckol Improves Glutamate-Induced Neurotoxicity through Modulation of Oxidative Stress-Mediated Mitochondrial Dysfunction in PC12 Cells

Stroke is a complex neurodegenerative disorder with a clinically high prevalence and mortality. Despite many efforts to protect against ischemic stroke, its incidence and related permanent disabilities continue to increase. In this study, we found that pretreatment with phlorofucofuroeckol (PFF), isolated from brown algae species, significantly increased cell viability in glutamate-stimulated P...

Sodium valproate ameliorates aluminum-induced oxidative stress and apoptosis of PC12 cells

Objective(s): According to recent studies, valproate shows some protection against oxidative stress (OS) induced by neurotoxins. Current investigation tried to determine the possible ameliorating effects of sodium valproate (SV) against aluminum (Al)-induced cell death, apoptosis, mitochondrial membrane potential (MMP), and OS in PC12 cells.Materials ...

Protective effects of nimodipine and lithium against aluminum-induced cell death and oxidative stress in PC12 cells

Objective(s): The role of aluminum (Al) in the pathogenesis of neurodegenerative diseases has been implicated in several studies. However, the exact mechanisms of cytotoxic effects of Al have not been elucidated yet. The aim of this study was to investigate the effect of L-type calcium channel antagonist, nimodipine (NM), and lithium chloride (LiCl) on Al-induced toxicity in PC12 cells. Materia...

The Hydroalcoholic Extract of Saffron Protects PC12 Cells against Aluminum-Induced Cell Death and Oxidative Stress in Vitro

Background: Aluminum (Al) exposure is among the environmental risk factors that may involve in the pathogenesis of neurodegenerative diseases. Oxidative stress has a critical role in the Al-induced toxicity. Saffron is a plant with potent radical scavenging and anti-oxidative properties. This investigation was designed to evaluate the possible protective effects of saffron extract (SE) on alumi...
